Serial & Large-Area Sectioning Service

Standard microscopy slides capture only a fraction of the biological picture. For researchers building 3D tissue atlases or analyzing whole organs, standard cutting techniques fall short.

Serial & Large-Area Sectioning service at Creatve Proteomics is designed for projects that demand scale and continuity. Whether you are reconstructing a mouse brain volume or profiling a whole human heart ventricle, we provide the technical artistry required to maintain spatial context across large distances and hundreds of sections.

Precision Serial Sectioning For 3D Reconstruction & Multi-Omics Layering

Serial sectioning refers to the continuous cutting of the same tissue sample along the same direction, yielding a series of sections that are highly spatially correlated. This strategy maintains good spatial continuity along the depth (Z-axis) of the tissue and is suitable for multi-layer information integration and three-dimensional structural analysis. We offer automated, high-precision serial sectioning for:

  • 3D Volumetric Reconstruction: Generating hundreds of consecutive sections with identical thickness to build digital 3D models of tumors, embryos, or brain structures.
  • Multi-Omics "Stacks": We create perfectly adjacent sections for different modalities, allowing you to layer disparate data sets on the same tissue block.
    > Section 1 (5µm): H&E Staining (Morphology)
    > Section 2 (10µm): Spatial Transcriptomics (Visium/Xenium/GeoMx)
    > Section 3 (10µm): Spatial Proteomics (IHC/IF)
    > Section 4 (12µm): Spatial Lipidomics (MALDI-MSI)

Applications:

  • Research on Tumor Microenvironment Heterogeneity
  • Mechanism-driven Spatial Multi-omics Integration
  • Longitudinal and High-resolution Analysis of Key Functional Regions

Serial Sectioning

Large-Area Sectioning for Whole-Organ Context on a Single Slide

Large-area sectioning emphasizes preserving complete and continuous tissue structures within a single section, enabling the comprehensive presentation of large-scale spatial information. This strategy is suitable for constructing spatial omics atlases of entire tissues or large regions. Our lab is equipped with specialized macro-cryostats and heavy-duty microtomes capable of processing "Jumbo" and "Super-Mega" cassettes.

  • Whole-Body Cryosectioning: Sagittal or coronal sections of whole mice or rats for drug distribution (PK/PD) studies.
  • Human Organ Mapping: Full cross-sections of human brain hemispheres, liver lobes, or cardiac ventricles.
  • Maximum Surface Area: We can process blocks up to 150mm x 100mm, mounted on large-format glass and conductive ITO slides compatible with select spatial scanners (e.g., Bruker MALDI, select slide scanners).

Applications:

  • Tumor center-periphery and tumor-normal tissue interface analysis
  • Spatial distribution studies of immune cells across the entire tissue scale
  • Construction of comprehensive spatial transcriptomics, spatial proteomics, and spatial metabolomics maps

Large-Area Sectioning

Why "The Cut" Matters for Spatial Omics

In large-area and serial sectioning, mechanical artifacts are the enemy of data. A fold in the tissue isn't just a visual blemish—it is a region where spatial data is lost or distorted.

Our histotechnologists use proprietary anti-roll systems and specialized tape-transfer methods (where applicable) to ensure:

  • Zero Folding: Essential for accurate image registration.
  • No Chattering: Smooth surfaces ensure laser/probe focus remains constant across the entire slide.
  • Uniform Adhesion: Preventing tissue detachment during the long incubation steps of spatial assays.

In practical research, serial sectioning and large-area sectioning can be flexibly combined. Large-area sectioning is used to obtain the global spatial pattern, while serial sectioning is used for in-depth multi-layer and multi-omics analysis of key regions. This combined strategy can ensure the integrity of macroscopic structures while achieving precise analysis at the microscopic mechanism level, providing high-quality data support for spatial omics research and translational medicine.

FAQs

Q:Can Serial Section and Large-Area Sectioning be combined in one project?

A:Yes. These two strategies are highly complementary. Large-area sectioning can be used to identify global spatial patterns, while serial sectioning allows detailed multi-layer and multi-omics analysis of selected regions of interest.

Q: Do large-area sections affect data quality or resolution?

A: Large-area sectioning preserves overall tissue architecture without compromising spatial resolution. However, it requires optimized sample preparation and imaging workflows to ensure uniform section quality and data consistency across the entire tissue area.
