Antibody Light and Heavy Chain Sequencing

Antibody Light and Heavy Chain Sequencing

Service Details

Introduction

Antibody (Ab) is an immunoglobular protein with a "Y"-shaped structure secreted by effector B cells (effector lymphoid B cells). Its structure is basically the same, consisting of two heavy chains (Heavy, abbreviated as H) and two Light chain (Light, abbreviated as L). The stem of the "Y" is called the Fc segment and consists of the -COOH group terminal halves of the two heavy chain polypeptide chains. The Fc segments of the two heavy chains are linked by one or several disulfide bonds. The two forks of the "Y" are called Fab fragments, and each fork consists of a complete light chain and half of the heavy chain-NH base ends. The light chain includes a variable region and a constant region, and the variable region accounts for about 1/2 of the light chain. Heavy chains include variable and constant regions. Antibodies can be divided into different types according to the different heavy chains. such as, the heavy chains of mammalian Ig have five types: α, δ, ε, γ and μ. Correspondingly, mammalian Ig can be divided into IgA, IgD, IgE, IgG and IgM.

Our Services

Creative Proteomics has established a new monoclonal antibody sequencing workflow based on a de novo sequencing approach without the need for database searches on the basis of the mass spectrometry platform. Our experienced scientists use de novo sequencing engineering to ensure that target antibody sequences are extracted from tens of thousands of MS/MS spectral data to complete antibody sequencing, helping researchers obtain antibodies with 100% coverage and accurate antibody light and heavy chain amino acids sequence information.

At Creative Proteomics, our antibody sequencing services can provide high-quality support for the development of your monoclonal antibodies for their quality control testing needs. Our scientists will first degrade your antibody to generate peptide fragments through multiple enzymes with different cleavage to ensure that each region of the antibody can produce several overlapping peptide fragments. Each peptide was sequenced to generate a map of each fragment and analyzed in conjunction with bioinformatics PEAKS software.

Our Technical Advantages

  • Optimized high-resolution mass spectrometry workflow and state-of-the-art antibody de novo sequencing software ensure full-sequence amino acid sequencing of antibody light and heavy chains from N-terminal to C-terminal.
  • A variety of different enzymatic cleavage is used to achieve 100% full-sequence pieces back together of the light and heavy chains of antibody molecules.
  • Targeted data processing algorithms is formulated to ensure data integrity by combining with bioinformatics PEAKS and other mass spectrometry analysis software.
  • Multiple subtypes and different classes of antibodies can be sequenced.

Creative Proteomics provides professional antibody light and heavy chain sequencing services for global customers. Our team has successfully sequenced thousands of antibodies, including antibody reagents, therapeutics, and newly isolated antibodies, helping researchers decode their antibodies in a fraction of the time. Therefore, please contact us if your antibody sequence cannot be searched in online databases, or you are interested in our services.
