Loading Coarse Venom with Nanoparticles

Venom is the secretion of poisonous animals, which is a mixture of a large number of biologically active molecules. Although the venom can cause pathophysiological conditions, it may also be an effective therapeutic agent for many diseases. In detailed studies of the pathophysiological manifestations caused by the administration of venom, scientists discovered the rational application of venom in the development of therapeutically effective drugs. And nanomaterials can be used in combination with venom, as a drug carrier and drugs to bring some cost-effective drugs. Therefore, Creative Proteomics provides technical services for the application of nanotechnology in venom research for the development of new drugs and effective treatments.

Introduction

Some well-known poisonous animals, such as snakes, scorpions and spiders, have their venom as a molecular tool widely studied and have a strong potential for clinical structural optimization to be developed into new drugs. However, the non-specific cytotoxicity of the venom will negatively affect the cells around the target, and the venom will reduce its effectiveness before it reaches the target cell. A promising way to circumvent these problems is through nanotechnology. Nanoparticles have a high surface area, combined with appropriate functionalized venom molecules, which can be used as an effective treatment for certain diseases. The main goal of our service is to study nanoparticles used for drug delivery of venoms and help develop venoms into some feasible treatments.

Technical Route

The venom material can be provided by the customer or directly by us. The other test materials are medical grade and analytical grade and used as-is. The specific technical route is shown in the figure below.

  • Nanoparticle preparation. The types of nanoparticles we prepare include but are not limited to chitosan, nano-gold, nano-silver, magnetic and supermagnetic nanoparticles, dendrimers, etc. The prepared particles are lyophilized, weighed and stored. According to the influence of different parameters, including venom concentration, nanometer particle size, etc., the relevant preparation process will be changed accordingly.
  • Venom loading. The conjugation of the venom and the nanoparticles is mainly achieved by the adsorption method.
  • System characterization. The morphology and structure of the nanoparticles were characterized, and the percentage of venom loaded in the system was measured to evaluate the whole. SEM and FTIR are used to determine the morphology and size of nanoparticles, and Zetasizer is used to measure the size, size distribution and zeta potential of the particles. The venom-laden nanoparticles were removed, and the free venom content in the supernatant was determined.
