Introduce of FcγRs
FcγRs (Fc-gamma receptors) are a family of cell surface receptors that play a crucial role in the immune system's response to various pathogens and diseases. These receptors are responsible for binding to the Fc region of immunoglobulins (antibodies) and mediating several immune functions, such as phagocytosis and antibody-dependent cell-mediated cytotoxicity. Understanding the affinity of FcγRs is essential in the context of antibody-based therapeutics and immunotherapy. - Surface plasmon resonance (SPR)
Surface plasmon resonance is a label-free, real-time, and high-throughput technique used for studying molecular interactions, including protein-protein interactions. Creative Proteomics utilizes surface plasmon resonance for Fc receptor binding assays. Our SPR-based affinity testing service for FcγRs enables researchers to accurately measure the binding affinity and kinetics between FcγRs and antibodies. This information helps in the identification and selection of antibodies for various therapeutic applications.
Flow cytometry allows for the quantitative measurement of antibody drug affinity for FcγRs. In this assay, we incubate cell populations expressing FcγRs with different concentrations of antibody drug. The cells are then stained with a fluorescent tag specific for the antibody drug, allowing for the detection and quantification of bound antibodies. By analyzing the fluorescence intensity of the stained cells, we can help our clients determine the affinity of the antibody for FcγRs.
- Enzyme-linked immunosorbent assays (ELISA)
The enzyme-linked immunosorbent assay (ELISA) is a commonly used method for the determination of antibody-antigen interactions. At Creative Proteomics, we immobilize FcγR proteins on solid surfaces such as microtiter plates. After immobilizing the FcγR protein, different concentrations of antibody drugs are added to the plate to bind to the FcγR. After washing away unbound antibody, a secondary antibody conjugated to an enzyme, such as horseradish peroxidase (HRP) or alkaline phosphatase (AP), is added. This secondary antibody recognizes and binds to the Fc region of the antibody drug, enabling detection of the bound antibody. By measuring the absorbance or fluorescence of the product, we can help our clients determine the binding affinity of the antibody drug to the FcγR. The affinity is usually expressed as a dissociation constant (Kd), which represents the concentration of the antibody drug to the FcγR.
