 We offer a complete 2D gel electrophoresis service using cutting-edge equipments from Bio-Rad and GE Healthcare. We determine relative quantitation of proteins in 2D gels that are stained with SYPRO® Ruby and CyDyeTM DIGE Fluors. DIGE overcomes the reproducibility challenges of conventional 2D gel electrophoresis by running the control and experimental samples in the same gel. Up to three mass and charge-matched, spectrally resolvable fluorescent dyes (Cy2, Cy3 and Cy5) are utilized to label a control and two experimental protein samples in vitro prior to DIGE. Each sample is then imaged separately but perfectly overlaid, avoiding the challenge of spot alignment. Thus the confidence and margin of error during sample difference analysis are substantially improved. The difference as small as 1.5-fold changes can be detected for large volume spots.
