This
technique takes mass spectral snapshots of intact
tissue slices, revealing how proteins and peptides
are spatially distributed within a given sample.
Mass spectrometry is an attractive proposition for
imaging, as a large range of different compounds can
be detected simultaneously. Direct tissue analysis
using MALDI-MS allows the generation of profiles
while maintaining the integrity of the tissue,
displaying cellular localizations and avoiding
tedious extraction and purification steps. It is
relatively cheap compared to many other imaging
techniques, and it does not require any labeling of
interesting compounds.
Bruker
Daltonics FLEX series (UltraFlex III
TOF/TOF and Autoflex) and ABI 4800
are used for MALDI Imaging service
in our lab. Tissues excised with
various glass slides adapt for
Bruker and ABI MALDI-MS analysis.
BioMAP is used for image analysis.
